If there are several amenitys in a building with several entrances, is there a way to describe which amenity can be reached by which entrance. e.g. a Building with two entrances. One leads to flats and the other one is only for the shop in the front.

You could map the building indoors (two ways, each leading from an entrance to one amenity).

I could also think(!, I never saw this) of a relation assigning the different entrances to objects.

For the simple case you could just put all the amenity tags and the entrance tag on the same node on the building outline. But this doesn't solve the general problem. There could be three entrances on different sides of the building of which two lead to the amenity and one doesn't. In that case see aseerel's answer for a possible solution.
